Abstract
New applications for the use of distinguishable particulate labels available in a variety of hues and sized in the submicron range are described. These applications include profiling of cellular components, obtaining secretion patterns, identifying a multiplicity of components in chromatographic or electrophoretic techniques and identification of desired immunoglobulin secreting cells.
Claims4
A method to identify and recover cells contained within a cell sample that secrete an immunoglobulin of desired specificity and affinity which method comprises:
- (a) providing onto a sample surface a multiplicity of cell samples, each of which cell samples contains cells that secrete immunoglobulins onto the sample surface, said surface optionally comprising a capture reagent for immunoglobulins, wherein each cell sample is dispensed onto a specified location on the sample surface to which the cells secrete the immunoglobulins;
- (b) probing said locations on the sample surface with a multiplicity of epitopes and/or antigens each labeled with a distinguishable particulate label wherein said multiplicity includes desired epitopes and/or antigens and undesired epitopes and/or antigens to which the immunoglobulins bind;
- (c) selecting any location of the surface which comprises immunoglobulins that bind to desired epitopes and/or antigens, but not to undesired epitopes and/or antigens;
- (d) identifying the cell sample having the cells which secrete immunoglobulins at the selected location;
- and
- (e) recovering the identified cell sample.
