Abstract
The invention relates to the field of drug delivery, in particular, to compounds and methods for the chemical modification of a proteinaceous channel to be used in pharmaceutical delivery vehicles for controlled and/or localized release of therapeutic molecules (e.g., small molecules, peptides, proteins or other macromolecules). Provided are drug delivery vehicles comprising a pH- and/or light-responsive channel protein.
Claims19
A delivery vehicle for controlled and/or localized release of therapeutic molecules, said delivery vehicle comprising a channel protein, the channel protein modified with a pH-responsive or light-responsive compound of the general formula Q-C or P-C,
- wherein Q is a chargeable group selected from the group consisting of Q1, Q2, and Q3 as depicted in the following,
- wherein R1, R2 and R3 are independently selected from the group consisting of H, methyl (Me), ethyl (Et), propyl (Pr), isopropyl (iPr), phenyl (Ph), and benzyl (Bn), wherein P is a photocleavable chargeable group selected from the group consisting of P1, P2, P3, P4, P5, P6, P7 and P8 as depicted in the following,
- wherein R1 and R2 are independently selected from the group of H, Me, Et, Pr, iPr, Ph and Bn and wherein R3═H or OMe, and
- wherein C is a coupling group selected from the group consisting of C1, C2, C3, C4, C5 and C6 as depicted in the following,
- wherein X═Br or I.
A delivery vehicle comprising a protein, the protein modified with a pH-responsive or light-responsive compound of the general formula Q-C or P-C,
- wherein Q is a chargeable group selected from the group consisting of Q1, Q2, and Q3 as depicted in the following,
- wherein R1, R2 and R3 are independently selected from the group consisting of H, methyl (Me), ethyl (Et), propyl (Pr), isopropyl (iPr), phenyl (Ph) and benzyl (Bn),
- wherein P is a photocleavable chargeable group selected from the group consisting of P1, P2, P3, P4, P5, P6, P7 and P8 as depicted in the following,
- wherein R1 and R2 are independently selected from the group consisting of H, Me, Et, Pr, iPr, Ph and Bn and wherein R3═H or OMe, and
- wherein C is a coupling group selected from the group consisting of C1, C2, C3, C4, C5 and C6 as depicted in the following,
- wherein X═Br or I.
A delivery vehicle comprising a protein, the protein modified with a compound of the general formula Q-C or P-C,
- wherein Q is selected from the group consisting of Q, Q2, and Q3 as depicted in the following,
- wherein R1, R2 and R3 are independently selected from the group consisting of H, methyl (Me), ethyl (Et), propyl (Pr), isopropyl (iPr), phenyl (Ph) and benzyl (Bn),
- wherein P is a photocleavable chargeable group selected from the group consisting of P1, P2, P3, P4, P5, P6, P7 and P8 as depicted in the following,
- wherein R1 and R2 are independently selected from the group consisting of H, Me, Et, Pr, iPr, Ph and Bn and wherein R3═H or OMe, and
- wherein C is a coupling group selected from the group consisting of C1, C2, C3, C4, C5 and C6 as depicted in the following,
- wherein X═Br or I.
