Abstract
Disclosed is a smooth muscle cell specific promoter, the SM22α gene promoter as well as the murine cDNA and genomic SM22α nucleic acid sequences. Also disclosed are methods of preventing restenosis following balloon angioplasty and methods of treating asthma based on inhibition of smooth muscle cell proliferation by expressing cell cycle control genes, or contraction inhibiting peptides in smooth muscle cells, under the control of the SM22α promoter.
Claims15
A method for promoting angiogenesis in a mammal comprising the step of providing to a smooth muscle cell in said mammal a nucleic acid construct comprising an SM22α promoter operably linked to a nucleotide sequence encoding a polypeptide or RNA competent to induce angiogenesis, said SM22α promoter having a maximum of 441 bases and comprising:
- (a) bases 900-1340 of SEQ ID NO:1 or
- (b) a sequence hybridizing with the complement of SEQ ID NO:1 under hybridizing conditions comprising 0.02M-0.15M sodium chloride at temperatures of 50° C. to 70° C., such that said nucleotide sequence encoding a polypeptide or RNA, when expressed in said smooth muscle cell, is competent to promote angiogenesis in said mammal.
A method for promoting angiogenesis in a mammal comprising the step of providing to a smooth muscle cell in said mammal a nucleic acid construct comprising an SM22α promoter operably linked to a nucleotide sequence encoding a polypeptide or RNA competent to induce angiogenesis, said promoter having a maximum of 441 bases and comprising:
- (a) an oligomer comprising SME1 (SEQ ID NO:21), SME2 (SEQ ID NO:23), SME3 (SEQ ID NO:25), SME4 (SEQ ID NO:27), SME5 (SEQ ID NO:31), or SME6 (SEQ ID NO:33), or
- (b) a sequence that hybridizes under hybridizing conditions comprising 0.02M-0.15M sodium chloride at temperatures of 50° C. to 70° C. with the complement of SME1 (SEQ ID NO:21), SME2 (SEQ ID NO:23), SME3 (SEQ ID NO:25), SME4 (SEQ ID NO:27), SME5 (SEQ ID NO:31), or SME6 (SEQ ID NO:33), such that said nucleotide sequence encoding said polypeptide or RNA, when expressed in said smooth muscle cell, is competent to promote angiogenesis in said mammal.
