Abstract
The present invention relates, in general, to a nucleoside base and, in particular, to a universal, photocleavable nucleoside base. The invention further relates to oligonucleotides comprising such a base.
Claims4
A compound of formula I
- wherein R1 is —H and R2 is —H, —OH or —OY, wherein Y is C1-4 alkyl,
- R3 and R4 are, independently, —H, a protecting group, or a mono-, di- or triphosphate, and
- X is O or S.
A compound comprising the moiety of formula II
- wherein R1 is —H and R2 is —H, —OH or —OY wherein Y is C1-4 alkyl, and
- X is O or S.
A method of synthesizing a nucleic acid susceptible to site specific cleavage comprising:
- reacting a compound of the formula
- wherein R1 is —H and R2 is —H, —OH or —OY, wherein Y is C1-4 alkyl, and X is O or S,
- with an oligonucleotide under conditions such that a nucleic acid comprising a moiety of the formula
- is produced, said nucleic acid being susceptible to site specific cleavage.
