Abstract
Novel human chemokine receptors, MCP-1RA and MCP-1RB, and processes for producing them are disclosed. The receptors, which are alternately spliced versions of MCP-1 receptor protein may be used in an assay to identify antagonists of MCP-1 which are therapeutically useful in the treatment of atherosclerosis and other diseases characterized by monocytic infiltrates.
Claims17
A heterologous polypeptide produced by expression of a heterologous nucleic acid in a host cell, wherein said polypeptide comprises an amino acid sequence identical to the sequence of the extracellular amino terminal domain of a mammalian MCP-1 receptor, wherein said receptor is encoded by a cDNA which is present in a mammalian library and which will hybridize under stringent conditions with a probe having the nucleotide sequence of the complement of the coding sequence shown in SEQ ID No. 1 or SEQ ID No. 3.
An isolated polypeptide comprising the amino acid sequence of the extracellular amino terminal domain of a mammalian MCP-1 receptor, wherein said receptor is encoded by a cDNA which is present in a mammalian library and which will hybridize under stringent conditions with a probe having the nucleotide sequence of the complement of the coding sequence shown in SEQ ID NO. 1 or SEQ ID No. 3.
An isolated polypeptide comprising the amino acid sequence of SEQ ID NO:
- 2 (FIG. 1) or SEQ ID NO:
- 4 (FIG. 2).
An isolated immunogenic polypeptide comprising a fragment of SEQ ID No:
- 2 or SEQ ID No:
- 4, wherein said fragment has the amino acid sequence of an extracellular domain of MCP-1RA, substantially as shown in FIGS. 4A and 4B.
