Abstract
A human gene has been discovered which is genetically altered in human tumor cells. The genetic alteration is gene amplification and leads to a corresponding increase in gene products. Detecting that the gene, designated hMDM2, has become amplified or detecting increased expression of gene products is diagnostic of tumorigenesis. Human MDM2 protein binds to human p53 and allows the cell to escape from p53-regulated growth. Methods of identifying compounds which interfere with the binding of human MDM2 protein to human p53 protein involved host cells which contain p-53-responsive reporter constructs and MDM2 polypeptides.
Claims4
A method for identifying compounds which interfere with the binding of human MDM2 to human p53, comprising the steps of:
- providing a cell which:
- (a) expresses an MDM2 polypeptide and a p53 polypeptide, wherein each of said polypeptides contains a sufficient portion of p53 or MDM2 proteins to bind to the other polypeptide;
- (b) contains a reporter construct whose expression is enhanced in the presence of the p53 polypeptide, said reporter construct comprising:
- (i) a DNA sequence which binds the p53 polypeptide;
- (ii) a promoter;
- and
- (iii) a reporter gene under the control of said promoter;
- and
- (c) expresses sufficient of said MDM2 polypeptide to inhibit expression of said reporter gene;
- contacting said cell with a compound to be tested for its capacity to inhibit binding of MDM2 and p53;
- determining the amount of expression of the reporter gene in the presence of said compound;
- wherein a compound which increases expression of the reporter gene is identified as a compound which inhibits the binding of MDM2 and p53.
A method of identifying compounds which interfere with the binding of human MDM2 to human p53, comprising:
- providing a cell which comprises three recombinant DNA constructs, said first construct encoding a first polypeptide fused to a sequence-specific DNA-binding domain, said second contact encoding a second polypeptide fused to a transcriptional activation domain, said third construct comprising a reporter gene downstream from a DNA element which is recognized by said sequence-specific DNA-binding domain, wherein said first polypeptide is an MDM2 polypeptide and said second protein is a p53 polypeptide, or said first polypeptide is a p53 polypeptide and said second polypeptide is an MDM2 polypeptide;
- wherein said polypeptides contain a sufficient portion of p53 and MDM2 proteins to bind to the other polypeptide;
- contacting said cell with a compound to be tested for its capacity to inhibit binding of p53 to MDM2;
- determining the amount of expression of the reporter gene in the presence of said compound, wherein a compound which decreases the expression of the reporter gene is a compound which inhibits the binding of p53 to MDM2.
A cell which:
- (a) expresses an MDM2 polypeptide and a p53 polypeptide, wherein each of said polypeptides contains a sufficient portion of p53 or MDM2 proteins to bind to the other polypeptide;
- (b) contains a reporter construct whose expression is enhanced in the presence of the p53 polypeptide, said reporter construct comprising:
- (i) a DNA sequence which binds the p53 polypeptide;
- (ii) a promoter;
- and
- (iii) a reporter gene under the control of said promoter;
- and
- (c) expresses sufficient of said MDM2 polypeptide to inhibit expression of said reporter gene.
A cell which comprises three recombinant DNA constructs, said first construct encoding a first polypeptide fused to a sequence-specific DNA-binding domain, said second construct encoding a second polypeptide fused to a transcriptional activation domain, said third construct comprising a reporter gene downstream from a DNA element which is recognized by said sequence-specific DNA-binding domain, wherein said first polypeptide is an MDM2 polypeptide and said second protein is a p53 polypeptide, or said first polypeptide is a p53 polypeptide and said second polypeptide is an MDM2 polypeptide;
- wherein said polypeptides contain a sufficient portion of p53 and MDM2 proteins to bind to the other polypeptide.
