Abstract
Pyridines, substituted by a quinuclidine and a substituent of low lipophilicity or a hydrocarbon substituent, their salts and prodrugs, and formulations thereof are useful in the treatment of neurodegenerative disorders such as dementia. The compounds can be synthesized by methods analogous to these known in the art.
Claims8
A pyridine compound of Formula I below:
- ##STR8## which is substituted on one of the ring carbon atoms with an R.sup.1 azabicyclic ring substituent selected from the group consisting of ##STR9## wherein the broken line represents a single or a, double bond;
- R.sup.3 and R.sup.4 independently represent hydrogen, C.sub.1-4 alkyl, halo, C.sub.1-4 alkoxy, hydroxy, carboxy or C.sub.1-4 alkoxycarbonyl;
- or R.sup.3 and R.sup.4 together represent carbonyl;
- R.sup.2, R.sup.11 and R.sup.12 are each independently selected from the group consisting of hydrogen, halo, --CF.sub.3, --OR.sup.6, --NR.sup.6 R.sup.7, --NHOR.sup.6, --NHNH.sub.2, --CN, COR.sup.8, and the hydrocarbon radicals:
- C.sub.1 -C.sub.15 alkyl, C.sub.2-15 alkenyl, C.sub.2-15 alkynyl, aryl, aralkyl, which in turn are optionally substituted by halo, OR.sup.6, CF.sub.3, NR.sup.6 R.sup.7, --NO.sub.2, keto, SR.sup.6, --SOR.sup.6, SO.sub.2 R.sup.6, CO.sub.2 R.sup.6, CONR.sup.6 R.sup.7, wherein R.sup.6 is selected from the group consisting of hydrogen, C.sub.1-6 alkyl, C.sub.2-6 alkenyl and C.sub.2-6 alkynyl;
- R.sup.7 is selected from the group consisting of hydrogen, C.sub.1-6 alkyl and --COCH.sub.3 and R.sup.8 is selected from the group consisting of --OR.sup.6 and --NR.sup.6 R.sup.7 ;
- and pharmaceutically acceptable salts and prodrugs thereof, provided that the compound is neither 3-hydroxy-3-(2-pyridinyl)-1-azabicyclo[2.2.2]octane nor 3-(2-pyridinyl)-1 -azabicyclo[2.2.2]oct-2 -ene.
4 more elements
A pharmaceutical composition comprising 3-(2-pyridinyl)-1azabicyclo[2.2.2]oct-2-ene in an effective amount as the active ingredient and a pharmaceutically acceptable carrier therefor.
