Abstract
In certain aspects, the invention provides methods and compositions for use in identifying a neutralizing status of a candidate antiviral agent. The invention also provides methods of determining one or more binding characteristics of glycan-binding agents.
Claims31
A method of identifying a virus-neutralization status of a candidate antiviral agent, the method comprising:
- (a) contacting a virion of a virus that has previously contracted an anti-virus antibody, with one or more preselected glycan-binding agents lectin molecules, wherein the contacting results in a virion/glycan-binding agent virion/lectin molecule complex;
- (b) contacting the virion/glycan-binding agent virion/lectin molecule complex with a candidate antiviral agent that is a candidate to block infection with the virus;
- and
- (c) determining one or more binding characteristics of the preselected glycan-binding agent(s) lectin molecule(s) with the antibody-contacted virion subsequent to the contacting with the candidate antiviral agent, wherein the determined one or more binding characteristics identifies the virus-neutralization status of the antiviral agent.
The method of claim 57 wherein the one or more binding characteristics are determined for binding of the one or more preselected lectin molecules with a virion protein.

The method of claim 57 wherein the determined binding characteristics comprise one or more of (a) the presence or absence of binding of the one or more preselected lectin molecule(s) with the virion protein;
- (b) a level of binding of the one or more preselected lectin molecules with the virion protein;
- (c) one or more location of the binding of the one or more preselected lectin molecules on the virion protein;
- (d) identity of the one or more preselected lectin molecules that bind the virion protein;
- and (e) identity of the one or more preselected lectin molecules that do not bind the virion protein.

The method of claim 57 wherein one or more of the preselected lectin molecules is detectably labeled.

The method of claim 57, wherein the virus is a SARS-CoV-2 virus, the anti-virus antibody is a purified antibody, and the one or more preselected lectin molecules contacting the virion protein comprise:
- MALI, RPL-αMan, RPL-Man2, PNA, RPL-Gal1, Jacalin, RPL-αGal, RPL-Sia1, RPL-Fuc1, DBA, and LTL, and wherein determining the presence of binding of the one or more preselected lectin molecules MALI, RPL-αMan, RPL-Man2, PNA, RPL-Gal1, Jacalin, RPL-αGal, RPL-Sia1, RPL-Fuc1, DBA, and LTL to the virion protein identifies the anti-virus antibody as a SARS-CoV-2-neutralizing antibody, and optionally further comprising administering the anti-viral antibody identified as neutralizing the virus to a subject as a treatment for a SARS-CoV-2 viral infection in the subject.

The method of claim 57, wherein the virus is a SARS-CoV-2 virus, the anti-viral antibody is obtained from a convalescent first subject patient serum sample, and the one or more preselected lectin molecules contacting the virion comprise:
- ConA, DBA, RCA I, PNA, SBA, UEA I, WGA, GSL I, LCA, PHA-E, PHA-L, PSA, LTL, DSL, ECL, GSL II, Jacalin, LEL, STL, VVL, AAL, MAL I, MAL II, SNL, RPL-αGal, RPL-Gal1, RPL-αMan, RPL-Man2, RPL-Sia1, and RPL-Fuc1, and wherein determining the presence of binding of the one or more preselected lectin molecules ConA, DBA, RCA I, PNA, SBA, UEA I, WGA, GSL I, LCA, PHA-E, PHA-L, PSA, LTL, DSL, ECL, GSL II, Jacalin, LEL, STL, VVL, AAL, MAL I, MAL II, SNL, RPL-αGal, RPL-Gal1, RPL-αMan, RPL-Man2, RPL-Sia1, and RPL-Fuc1 to the virion protein identifies the anti-virus antibody as a SARS-CoV-2-neutralizing antibody, optionally, further comprising administering the anti-viral antibody identified as neutralizing the virus to a second subject as a treatment for a SARS-CoV-2 viral infection in the second subject.
