Abstract
Systems and methods for generating viral vectors, including systems and methods for generating Dengue vaccine are described. Generally, a viral vector can comprise structural proteins from a non-dengue Orthoflavivirus species and a genetic code for nonstructural proteins from dengue virus or a different Orthoflavivirus species. Viral vectors can be produced to yield vaccines to stimulate immune response to the Orthoflavivirus species providing genetic code for nonstructural proteins.
Claims21
A viral vector system, comprising:
- one or more viral vector types, each viral vector type comprising:
- an outer layer comprising structural proteins of a non-dengue Orthoflavivirus species;
- and
- a nucleic acid for expressing one or more nonstructural proteins of dengue virus.
A method of yielding viral vector products, comprising:
- contacting a culture of packaging cells with:
- a nucleic acid comprising a first set of one or more expression constructs that encodes for one or more nonstructural proteins of dengue virus with a 5′-UTR a 3′-UTR, and a packaging signal, wherein the one or more nonstructural proteins are derived from native sequences of dengue virus;
- and
- a nucleic acid comprising a second set of one or more expression constructs that encodes for one or more structural proteins of a non-dengue Orthoflavivirus species, wherein the one or more structural proteins are derived from a native sequence of non-dengue Orthoflavivirus species.
A viral vector system, comprising:
- one or more viral vector types, each viral vector type comprising:
- an outer layer comprising structural proteins of Zika virus;
- and
- a nucleic acid for expressing one or more nonstructural proteins of a non-Zika Orthoflavivirus species.
