Abstract
The invention relates to the production, modification, and use of extracellular vesicles (EVs), particularly for the delivery of payloads of nucleic acids. The EVs can contain retroviral capsids and can be coated with polymers. The EVs can be treated with a protease to remove proteins on the outer surface of the EV to create shaved EVs. The shaved EVs can be coated with a polymer and used for the delivery of payloads of nucleic acids to cells.
Claims24
A mammalian-cell-derived extracellular vesicle (EV) that has been treated with a protease to remove proteins on the outer surface of the EV, wherein the surface of the EV is at least partially coated with at least one polymer selected from the group consisting of polyarginine, polylysine, polyethylene Imine (PEI), chitosan, poly(β-amino ester) (PBAE), PEI-co-polyhistidine copolymer, polylysine dendigraft/dendrimer/branched polymer, star shaped polyaminoacid based polymer, chondroitin sulfate, heparosan, poly glutamic acid (PGA), hyaluronic acid, dextran sulfate, polysarcosine, lipid-polyethyleneglycol (PEG), polycarboxybetaine, polysulfobetaine, and polyphosphorylcholine.

A method of modifying the outer surface of mammalian-cell-derived extracellular vesicles (EVs) comprising:
- a) purifying EVs from a mammalian cell;
- b) treating the EVs with a protease to remove proteins on the outer surface of the EVs;
- and
- c) purifying the treated EVs from the protease, wherein the surface of the EVs is at least partially coated with at least one polymer selected from the group consisting of polyarginine, polylysine, polyethylene Imine (PEI), chitosan, poly(β-amino ester) (PBAE), PEI-co-polyhistidine copolymer, polylysine dendigraft/dendrimer/branched polymer, star shaped polyaminoacid based polymer, chondroitin sulfate, heparosan, poly glutamic acid (PGA), hyaluronic acid, dextran sulfate, polysarcosine, lipid-polyethyleneglycol (PEG), polycarboxybetaine, polysulfobetaine, and polyphosphorylcholine.
