Abstract
The present invention provides a compound as represented by formula (I), and a pharmaceutical composition, and a preparation method therefor and a use thereof. The compound has a good inhibitory effect on KIF18A, can be used alone or in combination with microtubules to form a bonded complex to modulate a KIF18A protein for treatment of KIF18A-mediated conditions and/or diseases, such as tumors, and for preparation of a drug for such conditions or diseases.
Claims10
A compound represented by formula (I), a racemate, stereoisomer, tautomer, isotope-labeled counterpart, solvate, polymorph, pharmaceutically acceptable salt thereof or a prodrug compound thereof:
- wherein, A is selected from fused ring groups which are unsubstituted or optionally substituted by one, two or more Ra groups;
- the fused ring group comprises two, three or four rings independently selected from a saturated or partially unsaturated C3-14 carbon ring, a C6-14 aromatic ring, a 5- to 14-membered heteroaromatic ring, or a 3- to 14-membered heterocyclic ring;
- the Ra groups are each identical or different and are independently selected from H, OH, halogen, cyano, NH2, NO2, the following groups which are unsubstituted or optionally substituted by one, two or more Ra1 groups:
- C1-12 alkyl, C1-12 alkoxy, C3-12 cycloalkyl;
- or two Ra groups attached to the same ring carbon atom together with the carbon atom to which they are attached form a saturated or partially unsaturated C3-14 carbon ring;
- or, two non-adjacent Ra groups are linked at their end groups to form C1-3 alkylene;
- the Ra1 groups are each identical or different and are independently selected from H, OH, halogen, cyano, NH2, NO2, C1-12 alkyl, C1-12 alkoxy, C3-12 cycloalkyl;
- E is selected from the following groups which are unsubstituted or optionally substituted by one, two or more Re groups:
- —NH—S(═O)2—Re1, —S(═O)2—NH—Re2, —S(═O)(═NH)—Re3, —N(Re4)(Re5), 3- to 14-membered heterocyclic groups;
- the Re groups are each identical or different and are independently selected from OH, halogen, cyano, C1-12 alkyl, C1-12 alkoxy, halo-C1-12 alkyl, halo-C1-12 alkoxy, cyano-C1-12 alkyl, cyano-C1-12 alkoxy, —N(Re6)(Re7);
- the Re1, Re2, Re3, Re4, Re5, Re6, Re7 groups are each identical or different and are independently selected from H, C1-12 alkyl, hydroxy-C1-12 alkyl, halo-C1-12 alkyl, halo-C1-12 alkoxy, cyano-C1-12 alkyl, cyano-C1-12 alkoxy, C3-12 cycloalkyl, 3- to 14-membered heterocyclic groups, C1-12 alkoxy-C1-12 alkyl;
- M is selected from C3-12 cycloalkyl, C3-12 cycloalkenyl or nitrogen-containing 3- to 14-membered heterocyclic groups, which are unsubstituted or optionally substituted by one, two or more Rm groups;
- the Rm groups are each identical or different and are independently selected from H, halogen, cyano, C1-12 alkyl, halo-C1-12 alkyl, cyano-C1-12 alkyl, C1-12 alkoxy, cyano-C1-12 alkoxy;
- or, two Rm groups attached to the same ring carbon atom together with the carbon atom to which they are attached form a saturated or partially unsaturated C3-4 carbon ring;
- Y1, Y2, Y3 are identical or different and are independently selected from N or CH.
