Abstract
Aspects of the present disclosure include analyzing flow cytometers that exhibit reduced sample carryover. To reduce sample carryover, analyzing flow cytometers of embodiments of the invention are configured to bidirectionally transmit sheath fluid through a sample line of the analyzing flow cytometer. Also provided are methods of processing a sample using the disclosed analyzing flow cytometers.
Claims22
An analyzing flow cytometer comprising:
- a flow cell comprising:
- a sample line inlet;
- a sheath fluid injection port;
- and
- a waste fluid outlet;
- a sample line coupled to the sample line inlet and configured to receive a sample from a sample container;
- a sheath line coupled to the sheath fluid injection port and configured to transmit sheath fluid from a sheath fluid container to the sheath fluid injection port;
- a first pump fluidically coupled to the waste fluid outlet;
- a second pump fluidically coupled to the sheath line;
- and
- a controller configured to direct the first pump, the second pump, or both to bidirectionally transmit sheath fluid through the sample line.
