Abstract
Embodiments of the present invention encompass systems and methods for determining detection limits for various antibody-dye conjugates for flow cytometry. Exemplary techniques involve a linear superpositioning approach of spillover-induced enlargements of normally distributed measurement errors.
Claims21
A method for determining an optimized probe panel for differentiating populations of particles in a sample using a simulator module, the method comprising:
- receiving, via a user interface, user input corresponding to one or more parameters of a probe panel, the one or more parameters including a set of antibodies corresponding to an antigenic expression pattern;
- determining, via a simulator module, the optimized probe panel based on the user input and optionally one or more of the following:
- a flow cytometry device configuration and an antibody database, the determining comprising:
- running a simulation of probe panels via the simulator module that satisfy the user input and one or more of the flow cytometry device configuration and the antibody database;
- generating one or more optimized probe panels;
- and
- displaying at least one of the one or more optimized probe panels.
A system for determining an optimized probe panel for differentiating populations of particles in a sample, the system comprising:
- a user interface configured to receive user input corresponding to one or more parameters of a probe panel, the one or more parameters including a set of antibodies corresponding to an antigenic expression pattern;
- a simulator module in electronic communication with the user interface, the simulator module configured to determine the optimized probe panel based on the user input and one or more of the following:
- a flow cytometry device configuration and an antibody database, by:
- running a simulation of probe panels that satisfy the user input and one or more of the flow cytometry device configuration and the antibody database;
- generating one or more optimized probe panels;
- and
- a memory in electronic communication with the processor, the memory storing the antibody database module,
- wherein the simulator module is configured to produce graphs showing detection limits and population separations based on simulated probe panels.
A user interface for determining an optimized probe panel for differentiating populations of particles in a sample, the user interface comprising:
- a user input device configured to receive user input corresponding to one or more parameters of a probe panel, the one or more parameters including a set of antibodies corresponding to an antigenic expression pattern;
- and
- a user output device configured to display graphs generated by a simulator module showing detection limits and population separations based on simulated probe panels.
