Abstract
This disclosure concerns a recombinant protein comprising one or more repeating units, each of the one or more repeating units comprising a chemical cleavage site and a molecular weight polypeptide standard. The disclosure also concerns corresponding nucleic acids, vectors and host cells, a method of producing a plurality of molecular weight polypeptide standards and the resultant set of molecular weight standards and use thereof for calibrating a MALDI-TOF mass spectrometer.
Claims20
A recombinant protein comprising one or more repeating units, each of the one or more repeating units comprising a chemical cleavage site and a molecular weight polypeptide standard, wherein the chemical cleavage site is an amino acid sequence, the recombinant protein arranged according to formula I
- wherein
- brackets indicate a repeating unit of the one or more repeating units;
- PP is a molecular weight polypeptide standard;
- CCS is a chemical cleavage site;
- and
- n is an integer from 1 to 50.
A method of producing a plurality of molecular weight polypeptide standards, the method comprising:
- a) expressing a recombinant protein from a nucleic acid sequence encoding the recombinant protein, the recombinant protein comprising one or more repeating units, each of the one or more repeating units comprising a chemical cleavage site and a molecular weight polypeptide standard, wherein the chemical cleavage site is an amino acid sequence, the recombinant protein arranged according to formula I
- wherein
- brackets indicate a repeating unit of the one or more repeating units;
- PP is a molecular weight polypeptide standard;
- CCS is a chemical cleavage site;
- and
- n is an integer from 1 to 50;
- b) isolating the expressed recombinant protein;
- c) cleaving the chemical cleavage site or sites of the isolated recombinant protein using a chemical agent that recognizes the chemical cleavage site to produce a plurality of molecular weight polypeptide standards, wherein each molecular weight polypeptide standard has its amino acid sequence without any extraneous amino acids.
