Imidazodiazepinediones and methods of use thereof

A compound of Formula (I) or (II), or a tautomer or a pharmaceutically acceptable salt thereof,

• wherein
• A and A′ are CR;
• R is L-R1;
• each of L is independently selected from absent, CH2, O, SO2, or NR2;
• each of R1 is independently selected from optionally substituted alkyl, optionally substituted aryl, and optionally substituted heteroaryl, and when L is absent, R1 is additionally selected from H;
• each R2 is independently H, or alkyl;
• R3 is selected from optionally substituted alkyl, optionally substituted alkylene-OR2, optionally substituted cycloalkylene-OR2, optionally substituted alkylene-N(R7)2, optionally substituted cycloalkylene-N(R7)2, optionally substituted alkylene-C(O)N(R2)2, optionally substituted cycloalkylene-C(O)N(R2)2, optionally substituted alkylene-S(O)2N(R2)2, and optionally substituted cycloalkylene-S(O)2N(R2)2;
• R4 is selected from alkyl, optionally substituted alkylene-aryl, and optionally substituted alkylene-heteroaryl;
• each R5 is independently selected from H, N(R2)2, OR2;
• each R7 is independently selected from H, alkyl, (alkyl)C(O)—, (aryl)C(O)—, (alkyl)S(O)2—, and (aryl)S(O)2—;
• X is —C(O)—, CH2, CHR6, C(R6)2;
• each R6 is independently selected from H, alkyl, and optionally substituted alkylene-OH;
• X′ is —C(O)—, CH2, CHR3′, C(R3′)2, or X′ is taken together with R3 to form a 5- or 6-membered ring;
• each R3′ is independently selected from optionally substituted alkyl, optionally substituted alkylene-OR2, optionally substituted cycloalkylene-OR2, optionally substituted alkylene-N(R7)2, optionally substituted cycloalkylene-N(R7)2, optionally substituted alkylene-C(O)N(R2)2, optionally substituted cycloalkylene-C(O)N(R2)2, optionally substituted alkylene-S(O)2N(R2)2, and optionally substituted cycloalkylene-S(O)2N(R2)2;
• and
• Z is absent, CH2, CHR5, O, —NR2—, or —SO2—;
• provided that X and X′ are not both —C(O)—, and provided that R5 is H when Z is O, —NR2—, or —SO2—.
