Method of treatment or prophylaxis

A computer-implemented method for guiding treatment of an individual subject having a complex disorder, the method comprising:

• receiving genome-wide genetic variant data from the subject;
• selecting a plurality of groups of genetic variants associated with the complex disorder, wherein each group of variants is defined using a different statistical significance threshold;
• performing gene set association analysis on each of the plurality of groups of genetic variants to identify one or more biological pathways enriched for variants associated with the complex disorder;
• calculating, for the subject, a pharmagenic enrichment score (PES) for at least one of the identified biological pathways by aggregating genetic variants mapped to genes within the biological pathway;
• comparing the pharmagenic enrichment score for the subject to a reference distribution;
• and
• determining whether modulation of the biological pathway is likely to provide therapeutic benefit or lack thereof to the subject.
