Abstract
Current cell therapies and gene therapies used to increase or suppress the innate immune are often only temporarily effective due to clearance of reservoirs and the need to minimize risks of adverse systemic effects. Therefore, localized autonomous therapies are needed. Provided herein are transposon vectors and constructs for autonomously regulating expression and/or activity of proteins in inflammatory environments, and methods of their use.
Claims25
A transposon vector comprising a construct, the construct comprising:
- a gene encoding a therapeutic protein operably linked to a heterologous promoter;
- wherein the therapeutic protein inhibits an activator protein;
- and
- wherein the activator protein activates transcription of the gene encoding the therapeutic protein.
A construct comprising a sequence having at least 95% identity to SEQ ID NO:
- 1.
A construct comprising a sequence having at least 95% identity to SEQ ID NO:
- 3.
A construct comprising a sequence having at least 95% identity to SEQ ID NO:
- 5.
A construct comprising a sequence having at least 95% identity to SEQ ID NO:
- 13.
