-23. (canceled)
A polynucleotide comprising:
- (i) a first nucleic acid sequence encoding a chimeric antigen receptor (CAR), wherein the CAR comprises the amino acid sequence of SEQ ID NO.:
- 86;
- (ii) a second nucleic acid sequence at the 3′end of the first nucleic acid sequence, wherein the second nucleic acid sequence encodes a dominant negative TGFβ Receptor (DN TGFβR), wherein the DN TGFβR comprises from N- to C-terminus:
- an extracellular domain (ECD) from human TGFβ receptor II;
- a transmembrane domain (TMD) from human TGFβ receptor II, and
- an intracellular domain from human TGFβ receptor II lacking amino acid residues responsible for signaling and phosphorylation present in a wild type human TGFβ receptor;
- and
- (iii) a third nucleic acid sequence encoding a 2A peptide that links the first nucleic acid sequence encoding the chimeric antigen receptor to the second nucleic acid sequence encoding the DN TGFβR, wherein the third nucleic acid sequence comprises the sequence set forth in SEQ ID NO:
- 119.
6 more elements
A chimeric antigen receptor (CAR) comprising means for binding human GPC3, a transmembrane domain from a human protein, an intracellular costimulatory domain from a human protein, and an intracellular signaling domain from a human protein.
