Abstract
Disclosed are methods of assessing the ability of a candidate therapeutic agent to reverse, reduce or prevent intestinal injury by a potential toxic agent using a three-dimensional, engineered, bioprinted, biological intestinal tissue model. Also disclosed are methods of assessing the effect of an agent on intestinal function, the method comprising contacting the agent with a three-dimensional, engineered, bioprinted, biological intestinal tissue model.
Claims26
A three-dimensional, engineered, bioprinted, biological intestinal tissue model comprising:
- i. a layer of intestinal interstitial tissue comprising myofibroblasts;
- and
- ii. a layer of intestinal epithelial cells on the layer of intestinal interstitial tissue, to form the three-dimensional, engineered, biological intestinal tissue model,
- optionally wherein:
- at least one of the layer of intestinal interstitial tissue and layer of intestinal epithelial cells further comprises at least one type of immune cell,
- the layer of intestinal epithelial cells further comprises at least one stem cell population capable of differentiating,
- the intestinal tissue model further comprises a biocompatible membrane in contact with the intestinal tissue layer, or
- a combination thereof.

An intestinal tissue model comprising:
- (a) a polarized epithelial layer comprising intestinal epithelial cells,
- (b) an interstitial layer comprising intestinal myofibroblasts, and
- (c) a biocompatible membrane,
- wherein the layers are separated from one another by the biocompatible membrane, the basolateral surface of each of the layers is in contact with the biocompatible membrane, and the polarized epithelial layer is on top of the interstitial layer.
