Abstract
The present invention relates to microfluidic devices, such as cassettes for diagnostics, which allow for on-cassette processing including amplification by polymerase chain reaction (PCR), and systems and methods of using the same, wherein microfluidic cassette is pressurised prior to amplification of nucleic acid in said sample in an amplification/PCR section of the device.
Claims12
A method of amplifying nucleic acid from a sample, the method comprising:
- providing a microfluidic device comprising:
- a microfluidic channel having a polymerase chain reaction (PCR) section, the PCR section configured for temperature changes to occur therein;
- a fluidically isolatable portion comprising at least a portion of the microfluidic channel having the PCR section;
- at least one valve actuatable to fluidically isolate the fluidically isolatable portion from an external environment;
- and
- a plurality of positive displacement pumps;
- introducing a sample into the microfluidic device;
- fluidically isolating the fluidically isolatable portion to obtain a fluidically isolated portion;
- increasing a pressure within the fluidically isolated portion by simultaneously or concurrently compressing the plurality of positive displacement pumps to a first position to apply a first pressure to the fluidically isolated portion to increase the pressure in the fluidically isolated portion;
- carrying out polymerase chain reaction (PCR) amplification steps to amplify the nucleic acid within the sample;
- and
- further compressing one of the positive displacement pumps to a second position to move the sample within the microfluidic channel.
