Abstract
Methods are provided for the production of photoreceptor cells and photoreceptor progenitor cells from pluripotent stem cells. Additionally provided are compositions of photoreceptor cells and photoreceptor cells, as well as methods for the therapeutic use thereof. Exemplary methods may produce substantially pure cultures of photoreceptor cells and/or photoreceptor cells.

A preparation comprising a plurality of cell spheres, wherein at least 50% of the cells of the cell spheres are MASH1(+), CHX10(−), and a medium for maintaining the viability of the cells.
A preparation comprising a plurality of cell spheres, wherein at least 50% of the cells of the cell spheres are MASH1(+), CHX10(−), and a medium for maintaining the viability of the cells, wherein the cells of the preparation are differentiated in vitro from human pluripotent stem cells, wherein less than 1% of the cells of the preparation are human embryonic stem cells, induced pluripotent stem cells, eye field progenitor cells, retinal ganglion cells, mature photoreceptors, and/or amacrine cells, and wherein the MASH1(+), CHX10(−) cells:
- secrete one or more neuroprotective factors;
- have phagocytic activity;
- have the ability to phagocytose isolated photoreceptor outer segments, fluorescent bioparticles, or both photoreceptor outer segments and fluorescent bioparticles;
- or
- a combination thereof.
