Abstract
A stable long-acting growth hormone that uses recombinant human serum albumin and recombinant human growth hormone as ingredients and is formed by means of linker conjugation, as well as a preparation method therefor. An rHSA-hGH conjugate having a purity not lower than 95% is obtained after separation and purification.
Claims7
A polypeptide conjugate of human serum albumin and human growth hormone, wherein the polypeptide conjugate comprises a recombinant human serum albumin conjugated to a recombinant human growth hormone via a linker group,
- wherein the linker group comprises N-succinimidyl 6-maleimidohexanoate, and an amino of the recombinant human growth hormone is linked to an amino reactive group of the N-succinimidyl 6-maleimidohexanoate;
- wherein the recombinant human serum albumin is a plant-derived recombinant human serum albumin.
A method for preparing a polypeptide conjugate of human serum albumin and human growth hormone comprising the following steps in sequence:
- (a) mixing a recombinant human growth hormone with an activated N-succinimidyl 6-maleimidohexanoate under conditions that allow an amino of the recombinant human growth hormone to be linked to an amino reactive group of the N-succinimidyl 6-maleimidohexanoate, thereby obtaining a recombinant human growth hormone-EMCS intermediate conjugate product;
- (b) mixing the recombinant human growth hormone-EMCS intermediate conjugate product obtained in the step 1) with a plant-derived recombinant human serum albumin, thereby obtaining a human serum albumin-human growth hormone final conjugate product;
- and optionally further comprising:
- (c) purifying the obtained final conjugate product to obtain a purified preparation of the polypeptide conjugate of human serum albumin and human growth hormone.
