Abstract
A synthetic DNA molecule comprising one segment encoding a tumor neoantigen or an epitope from an infectious agent under the control of a promoter for the transcription into a corresponding RNA molecule and a segment for the translation of the translated RNA molecule into a peptide.
Claims15
A synthetic DNA molecule, comprising:
- a promoter configured for transcription of the synthetic DNA molecule into a corresponding RNA molecule;
- a segment configured for translation, by a Eukaryotic cell, of the corresponding RNA molecule into a peptide and/or configured for improvement of stability of the corresponding RNA molecule within the Eukaryotic cell;
- an Open Reading Frame comprising an epitope;
- and
- a 3′ Poly A tail segment,
- wherein the segment is configured for the translation to occur at the 5′ of the Open Reading Frame and/or the segment is configured for improvement of the stability of the corresponding RNA molecule at the 3′ of the Open Reading Frame,
- wherein the 3′ Poly A tail segment comprises at least 15 consecutive adenosine residues,
- wherein the epitope, when translated, comprises a size between 7 and 50 amino acids, and
- wherein the Open Reading Frame comprises a size between 21 and 500 nucleotides.
