Abstract
Methods, systems, and kits for sequencing a template nucleic acid molecule using a modified nucleotide molecule including a peptide spacer are provided. In some aspects, the modified nucleotide molecules are used in a sequencing reaction, such as an in situ sequencing reaction.
Claims24
A peptide-modified nucleotide having the structure:
- wherein N is a nucleotide comprising a 3′ blocked sugar and a base, L is a linker, P is a peptide comprising a charged residue, and D is a dye, and
- wherein the linker is attached to the base of the nucleotide.
A composition comprising a plurality of peptide-modified nucleotides, comprising:
- a first peptide-modified nucleotide molecule having a first base type and a second peptide-modified nucleotide molecule having a second base type, wherein the first and the second peptide-modified nucleotide molecules each have the structure N-L-P-D, wherein:
- N is a nucleotide molecule comprising a 3′ blocked sugar and a base, L is a linker, P is a peptide, and D is a dye.
A method for sequencing a template nucleic acid molecule comprising:
- (a) contacting a priming strand bound to a template nucleic acid molecule with a polymerase and a first plurality of nucleotide molecules comprising a first peptide-modified nucleotide having the structure N-L-P-D, wherein:
- N is a nucleotide molecule comprising a 3′ blocked sugar and a base, L is a linker, P is a peptide, and D is a dye, thereby incorporating the peptide-modified nucleotide into a 3′ terminus of the priming strand;
- and
- (b) detecting a presence of the first peptide-modified nucleotide in the priming strand to identify a complementary nucleotide in the template nucleic acid molecule.
1 more element
A kit for sequencing a template nucleic acid molecule comprising:
- a plurality of peptide-modified nucleotide molecule having the structure N-L-P-D, wherein:
- N is a nucleotide molecule comprising a 3′ blocked sugar and a base, L is a linker, P is a peptide, and D is a dye;
- and a polymerase.
