Abstract
The invention relates to compositions and methods for the preparation, manufacture, and therapeutic use of ribonucleic acid vaccines comprising polynucleotide molecules encoding one or more antigens.
Claims20
A vaccine comprising a messenger ribonucleic acid (mRNA) formulated in a lipid nanoparticle, wherein the mRNA in the vaccine comprises an open reading frame encoding an influenza hemagglutinin protein, wherein the open reading frame comprises nucleosides consisting of N1-methylpseudouridine, adenosine, guanosine, and cytidine, and wherein the lipid nanoparticle comprises an ionizable cationic lipid, a neutral lipid, a sterol, and a PEG-modified lipid.
The vaccine of 8, wherein the neutral lipid is DSPC and the sterol is a cholesterol.
The vaccine of 9, wherein the neutral lipid is DSPC and the sterol is a cholesterol.
A method of vaccinating a human subject against influenza, the method comprising administering to the human subject a dose of a vaccine comprising a messenger ribonucleic acid (mRNA) formulated in a lipid nanoparticle, wherein the mRNA comprises an open reading frame encoding an influenza hemagglutinin protein, wherein the open reading frame comprises nucleosides consisting of N1-methyl-pseudouridine, adenosine, guanosine, and cytidine, wherein the lipid nanoparticle comprises an ionizable cationic lipid, a neutral lipid, a cholesterol, and a polyethylene glycol (PEG)-modified lipid, and wherein the vaccine is administered intramuscularly.
