Abstract
The present invention is in the field of in vitro assays, particularly to detect pyrogens. The invention provides improved methods for animal-free approaches by providing an improved monocyte activation test. The improved test allows for reduced consumption of reagents and for improved throughput.

Method for detecting a pyrogen in a sample, the method comprising the steps of:
- i) providing one or more samples;
- ii) contacting the sample with peripheral blood mononuclear cells (PBMCs) in an incubation medium;
- and
- iii) determining the response of the PBMCs,
- wherein the incubation medium has a volume of at most 175 μL per sample.
