Abstract
Hydrogels comprising a macromolecular matrix and water may be used for aesthetic fillers, for example, dermal fillers. The macromolecular matrix may include a crosslinked combination of hyaluronic acid and collagen.
Claims22
A composition comprising about 6 mg/ml to about 20 mg/ml of a hyaluronic acid and about 3 mg/ml to about 12 mg/ml of a collagen, wherein the hyaluronic acid is crosslinked with the collagen via a plurality of crosslink units, and wherein at least a portion of the crosslink units comprise an amide bond.
A method of producing a crosslinked macromolecular matrix comprising:
- dissolving hyaluronic acid and a collagen in an aqueous solution to form an aqueous pre-reaction solution, wherein the aqueous pre-reaction solution further comprises a salt or has a pH less than 4;
- modifying the aqueous pre-reaction solution to form a crosslinking reaction mixture comprising:
- the hyaluronic acid;
- the collagen;
- a water-soluble carbodiimide;
- N-hydroxysuccinimide or N-hydroxysulfosuccinimide;
- and
- allowing the crosslinking reaction mixture to react so as to crosslink the hyaluronic acid with the collagen and form a crosslinked macromolecular matrix,
- wherein the crosslinking reaction mixture has a higher pH than the aqueous pre-reaction solution.
