Abstract
Methods of determining the risk of developing adjudicated arthropathy (AA) and/or progression to total joint replacement (TJR) and determining the treatment response to an NGF antagonist or NSAID treatment in a subject by determination of an OA proteomic risk score for the subject are presented herein.
Claims26
A method of determining the risk of a subject for developing adjudicated arthropathy (AA), the method comprising:
- determining or having determined the subject's osteoarthritis (OA) proteomic risk score, wherein the OA proteomic risk score comprises a weighted aggregate of the expression level or circulating level of one or more proteins associated with OA;
- wherein:
- when the subject's OA proteomic risk score is greater than or equal to a threshold OA proteomic risk score, the subject has an increased risk of developing AA;
- and
- when the subject's OA proteomic risk score is less than a threshold OA proteomic risk score, the subject has a decreased risk of developing AA.

A method of determining the risk of a subject having adjudicated arthritis (AA) that progresses to total joint replacement (TJR), the method comprising:
- determining or having determined the subject's osteoarthritis (OA) proteomic risk score, wherein the OA proteomic risk score comprises a weighted aggregate of the expression level or circulating level of one or more proteins associated with OA;
- wherein:
- when the subject's OA proteomic risk score is greater than or equal to a threshold OA proteomic risk score, the subject has an increased risk of progressing to TJR;
- and
- when the subject's OA proteomic risk score is less than a threshold OA proteomic risk score, the subject has a decreased risk of progressing to TJR.

A method of determining the treatment response of a subject having osteoarthritis (OA) treated with a Nerve Growth Factor (NGF) antagonist, the method comprising:
- determining or having determined the subject's OA proteomic risk score, wherein the OA proteomic risk score comprises a weighted aggregate of the expression level or circulating level of one or more proteins associated with OA;
- wherein:
- when the subject's OA proteomic risk score is greater than or equal to a threshold OA proteomic risk score, the subject has less response to an NGF antagonist;
- and
- when the subject's OA proteomic risk score is less than a threshold OA proteomic risk score, the subject has an increased response to an NGF antagonist.

A method of determining the treatment response (pain and functional improvement) of a subject having osteoarthritis (OA), the method comprising:
- determining or having determined the subject's OA proteomic risk score, wherein the OA proteomic risk score comprises a weighted aggregate of the expression level or circulating level of one or more proteins associated with OA;
- wherein:
- when the subject's OA proteomic risk score is greater than or equal to the first tertile of a threshold OA proteomic risk score, the subject has less treatment response to fasinumab 1q8w;
- and
- when the subject's OA proteomic risk score is less than the first tertile of a threshold OA proteomic risk score, the subject has an increase in treatment response to fasinumab 1q8w.
