Abstract
Provided are compounds of the Formula (I): or pharmaceutically acceptable salts thereof, which are useful for the inhibition of KIF18A and in the treatment of a variety of KIF18A mediated conditions or diseases, such as cancer.
Claims82
A compound represented by Formula (I)
- or a pharmaceutically acceptable salt thereof, wherein:
- X1 and X2 are each independently CR5 or N, and X3 is CR4 or N;
- Ring A is phenyl, 6-membered heteroaryl, 6,5-bicyclic heteroaryl, or 4- to 10-membered monocyclic or bicyclic heterocyclyl;
- Z is *—NHC(O)— or *—C(O)NH—, wherein *- represents the attachment to ring A;
- o is an integer from 0 to 3;
- R1 is C1-6alkyl, C3-6cycloalkyl, 3- to 6-membered monocyclic heterocyclyl, ORO1a, SO2R1a, NRN1aSO2R1a, NRN1aRN1b, —C(O)R1a, halo, cyano, wherein the C1-6alkyl, C3-6cycloalkyl and 3- to 6-membered monocyclic heterocyclyl are each optionally substituted with one or more R1b;
- R1a is C1-6alkyl, NRN1aRN1b, ORO1a, C3-6cycloalkyl, or 3- to 6-membered monocyclic heterocyclyl, wherein the C1-6alkyl, C3-6cycloalkyl, and 3- to 6-membered monocyclic heterocyclyl are each optionally substituted with one or more R1b;
- each R1b is independently selected from halo, cyano, hydroxy, C1-6alkyl, C1-6haloalkyl, C1-6alkoxy, and C1-6haloalkoxy;
- or two R1b, together with the atom or atoms to which they are attached, form C3-6cycloalkyl;
- RN1a and RN1b are each independently selected from H and C1-6alkyl, wherein the C1-6alkyl is optionally substituted with one or more R1b;
- RO1a is H or C1-6alkyl, wherein the C1-6alkyl is optionally substituted with one or more R1b;
- R2 is H, C1-6alkyl, SO2R2a, NRN2aSO2R2a, ORO2a, S(O)(NRN2c)R2a, halo, cyano, —C(O)R2a, or NRN2aRN2b, wherein the C1-6alkyl is optionally substituted with 1 or more R2b;
- R2a is C1-6alkyl, NRN2aRN2b, ORO2a, C3-6cycloalkyl, or 3- to 6-membered monocyclic heterocyclyl, wherein the C1-6alkyl, C3-6cycloalkyl, and heterocyclyl are each optionally substituted with 1 or more R2b;
- each R2b is independently selected from C1-6alkyl, halo, hydroxy, C1-6alkoxy, C1-6haloalkoxy, —N(RN2c)2, and —C(O)OC1-6alkyl;
- RN2a and RN2b are each independently selected from H and C1-6alkyl, wherein the C1-6alkyl is optionally substituted with one or more R2b;
- each RN2c is independently H, C1-3alkyl, —C(O)(C1-3alkyl);
- RO2a is H or C1-6alkyl, wherein the C1-6alkyl is optionally substituted with 1 or more halo, hydroxy, C1-6alkoxy, or C1-6haloalkoxy;
- R3 is C3-6cycloalkyl, phenyl, or 3- to 6-membered monocyclic heterocyclyl, wherein the 3- to 6-membered monocyclic heterocyclyl is optionally substituted with 1 or more R3a;
- each R3a is independently selected from halo, C1-6haloalkyl, or C1-6alkyl;
- or two R3a, together with the atom or atoms to which they are attached, form C3-6cycloalkyl substituted with 1 or more R3b;
- each R3b is independently selected from H, halo, C1-6alkoxy, and C1-6alkyl optionally substituted with one or more halo, OH, or C1-3alkoxy;
- R4 is H, C1-6alkyl, C1-6haloalkyl, or halo;
- R5 is H, halo, or C1-6alkyl;
- each R6 is independently C1-6alkyl, C1-6haloalkyl, or halo;
- provided that if ring A is phenyl or 6-membered heteroaryl, then R3 is
- where j is 0 or 1.
