A method for delivering a nucleic acid into an organ of a subject in need thereof, wherein the method comprises orally administering to the subject a pharmaceutical composition comprising a mixture of the nucleic acid and a lipid, and:
- wherein the organ is heart, spleen, kidney, or intestine;
- wherein the mixture of the nucleic acid and the lipid is prepared by a process comprising:
- mixing an organic solvent solution of the lipid with an aqueous solution of the nucleic acid;
- and
- heating the resulted mixture at about 80° C. to about 100° C. for about 15 to about 30 minutes to obtain the mixture of the nucleic acid and the lipid;
- and
- wherein the lipid is a compound of the formula I:
- wherein:
- L1 is —C(O)O—CH2—;
- A is C1-20 alkyl or C2-20 alkenyl;
- L2 is —C(O)O—;
- B is C1-20 alkyl or C2-20 alkenyl;
- L3 is —CH2—OC(O)—, and Q is C1-20 alkyl or C2-20 alkenyl;
- or L3 is —CH2—, and Q is —OH;
- with respect to the divalent groups L1 and L2, the dash “-” on the left side is linked to the groups A and B, respectively, and the dash “-” on the right side is linked to the central carbon atom;
- and
- with respect to the divalent group L3, the dash “-” on the left side is linked to the central carbon atom, and the dash “-” on the right side is linked to the group Q.
A compound of formula I:
- wherein L1, L2, and L3 are each independently absent or selected from the group consisting of —C(O)O—CH2—, —CH(OH)—, —C(O)—NH—CH2—, —CH2—OC(O)—, —CH2—NH—C(O)—, —C(O)O—, —C(O)NH—, —OC(O)—, —NH—C(O)—, —CH2—,
- with the proviso that at most two of L1, L2, and L3 are absent;
- with respect to the divalent groups L1 and L2, the dash “-” on the left side is linked to the groups A and B, respectively, and the dash “-” on the right side is linked to the central carbon atom;
- with respect to the divalent group L3, the dash “-” on the left side is linked to the central carbon atom, and the dash “-” on the right side is linked to the group Q;
- A, B and Q are each independently selected from the group consisting of H, —OH, C1-20 alkyl, C2-20 alkenyl, C1-20 heteroalkyl, C2-20 heteroalkenyl, —NH2, and —NR3+, R is H or C1-6 alkyl;
- and
- n is an integer 0, 1, 2, 3 or 4.
