Abstract
The subject matter described here relates to bispecific antibodies capable of modulating T-cell activity, wherein the two arms of the bispecific antibody are against CD3 and SLAMF6, against CD45 and SLAMF6, or against CD43 and SLAMF6.
Claims35
A bispecific antibody or a fragment thereof, comprising:
- a first arm comprising a first variable heavy chain domain and a first variable light chain domain, wherein a portion of the first arm is capable of binding to a portion of a CD3 protein, a portion of a CD45 protein, or a portion of a CD43 protein;
- and
- a second arm comprising a second variable heavy chain domain and a second variable light chain domain, wherein a portion of the second arm is capable of binding to a portion of a SLAMF6 protein.
A tetravalent bispecific antibody or a fragment thereof, comprising:
- (I) a first fragment antigen-binding (Fab) region, comprising:
- a first arm comprising a first variable heavy chain domain and a first variable light chain domain, wherein a portion of the first arm is capable of binding to a portion of a CD3 protein, a portion of a CD45 protein, or a portion of a CD43 protein;
- and
- a second arm comprising a second variable heavy chain domain and a second variable light chain domain, wherein a portion of the second arm is capable of binding to a portion of a CD3 protein, a portion of a CD45 protein, or a portion of a CD43 protein;
- (II) a first fragment, crystallizable (Fc) region;
- (III) a second Fab region, comprising:
- a third arm comprising a first variable heavy chain domain and a first variable light chain domain, wherein a portion of the first arm is capable of binding to a portion of a SLAMF6 protein;
- and
- a fourth arm comprising a second variable heavy chain domain and a second variable light chain domain, wherein a portion of the second arm is capable of binding to a portion of a SLAMF6 protein;
- and
- (IV) a second Fc region, wherein the first and second Fc regions associated together.
A kit for generating a bispecific antibody or fragment thereof, the kit comprising:
- a first vector comprising a polynucleotide sequence encoding a first arm of the bispecific antibody or fragment thereof, wherein a portion of the first arm is capable of binding to a portion of a CD3, protein, a portion of a CD45 protein, or a portion of a CD43 protein;
- and
- a second vector comprising a polynucleotide sequence encoding a second arm of the bispecific antibody or fragment thereof, wherein a portion of the second arm is capable of binding to a portion of a SLAMF6 protein.
One or more host cells comprising:
- a first vector comprising a polynucleotide sequence encoding a first arm of a bispecific antibody or fragment thereof, wherein a portion of the first arm is capable of binding to a portion of a CD3 protein, a portion of a CD45 protein, or a portion of a CD43 protein;
- and
- a second vector comprising a polynucleotide sequence encoding a second arm of the bispecific antibody or fragment thereof, wherein a portion of the second arm is capable of binding to a portion of a SLAMF6 protein.
A method of making a bispecific antibody or fragment thereof comprising:
- culturing the one or more host cells under conditions suitable for an expression of the one or more vectors;
- and
- recovering the bispecific antibody or fragment thereof.
A composition comprising:
- a first vector comprising a polynucleotide sequence encoding a first arm of the bispecific antibody or fragment thereof, wherein a portion of the first arm is capable of binding to a portion of a CD3 protein, a portion of a CD45 protein, or a portion of a CD43 protein;
- and
- a second vector comprising a polynucleotide sequence encoding a second arm of the bispecific antibody or fragment thereof, wherein a portion of the second arm is capable of binding to a portion of a SLAMF6 protein.
A means for binding:
- a portion of a CD3 protein, a portion of a CD45 protein, or a portion of a CD43 protein;
- and
- a portion of a SLAMF6 protein.
