Abstract
A substituted fused ring compound represented by formula (I), a pharmaceutical composition thereof, a preparation method therefor and the uses thereof are provided. The compound has a good LRRK2 kinase regulation/inhibition effect, and thus can be used for treating symptoms and diseases related to LRRK2 kinase activity, and can be used for preparing drugs for treating such symptoms and diseases.
Claims12
A compound represented by formula (I), or a racemate, a stereoisomer, a tautomer, an isotopically labeled compound, an N-oxide, a hydrate, a solvate, a polymorph, a metabolite, a pharmaceutically acceptable salt, a pharmaceutically acceptable ester, or a prodrug compound thereof:
- wherein,
- W is selected from N or O;
- R1 is selected from the following groups unsubstituted or substituted with one, two, or more Ra:
- C1-10 alkyl, C3-10 cycloalkyl, and 3- to 10-membered heterocyclyl, wherein each Ra is identical or different and is independently selected from halogen, hydroxyl, C1-10 alkyl, C1-10 alkyloxy, C3-10 cycloalkyl, C3-10 cycloalkyloxy, 3- to 10-membered heterocyclyl, and 3- to 10-membered heterocyclyloxy;
- or alternatively, two Ra, together with the atom(s) to which they are attached, form the following groups unsubstituted or optionally substituted with one, two, or more Rc:
- C1-10 alkyl, C1-10 alkyloxy, C3-10 cycloalkyl, C3-10 cycloalkyloxy, 3- to 10-membered heterocyclyl, and 3- to 10-membered heterocyclyloxy;
- R2 is absent or selected from H or C1-10 alkyl, wherein, when W is O, R2 is absent;
- when W is N, R2 is selected from H or C1-10 alkyl;
- or alternatively, R1 and R2, together with W, form 3- to 10-membered heterocyclyl unsubstituted or optionally substituted with one, two, or more Rb, wherein each Rb is identical or different and is independently selected from halogen, hydroxyl, C1-10 alkyl, C1-10 alkyloxy, C3-10 cycloalkyl, C3-10 cycloalkyloxy, 3- to 10-membered heterocyclyl, and 3- to 10-membered heterocyclyloxy;
- or alternatively, two Rb, together with the atom(s) to which they are attached, form the following groups unsubstituted or optionally substituted with one, two, or more Rc:
- C1-10 alkyl, C1-10 alkyloxy, C3-10 cycloalkyl, C3-10 cycloalkyloxy, 3- to 10-membered heterocyclyl, and 3- to 10-membered heterocyclyloxy, wherein each Rc is identical or different and is independently selected from halogen, hydroxyl, C1-10 alkyl, C1-10 alkyloxy, C3-10 cycloalkyl, C3-10 cycloalkyloxy, 3- to 10-membered heterocyclyl, and 3- to 10-membered heterocyclyloxy;
- X is selected from 5- to 10-membered heteroaryl substituted with one, two, or more R3, wherein each R3 is identical or different and is independently selected from the following groups unsubstituted or substituted with one, two, or more R4:
- C1-10 alkyl, C3-10 cycloalkyl, 3- to 10-membered heterocyclyl, and 5- to 10-membered heteroaryl;
- or alternatively, two R3, together with the atom(s) to which they are attached, form the following groups unsubstituted or optionally substituted with one, two, or more R4:
- C3-10 cycloalkyl and 3- to 10-membered heterocyclyl;
- wherein each R4 is identical or different and is independently selected from halogen, hydroxyl, cyano, or the following groups unsubstituted or substituted with one, two, or more Rd:
- C1-10 alkyl, C1-10 alkylene, C3-10 cycloalkyl, 3- to 10-membered heterocyclyl, and 5- to 10-membered heteroaryl, wherein when R4 is selected from C1-6 alkylene, it means that the carbon atom in R4, together with the carbon atom to which R3 is attached, forms a double bond;
- each Rd is identical or different and is independently selected from halogen, hydroxyl, cyano, C1-6 alkyl, C1-6 alkyloxy, C3-10 cycloalkyl, C3-10 cycloalkyloxy, 3- to 10-membered heterocyclyl, 3- to 10-membered heterocyclyloxy, 5- to 10-membered heteroaryl, and 5- to 10-membered heteroaryloxy.
