Abstract
Methods and systems for analysis of image data generated from various reference points. Particularly, the methods and systems provided are useful for real time analysis of image and sequence data generated during DNA sequencing methodologies.
Claims30
A method for analysis of image data generated during nucleic acid sequencing:
- obtaining fluorescent emission signals from nucleic acid sites on a flow cell, wherein a nucleic acid site comprises multiple copies of a nucleic acid, and wherein the fluorescent emission signals are indicative of nucleotides present at the nucleic acid site;
- identifying candidate site locations on the flow cell from the fluorescent emission signals, the fluorescent emission signals detected by different detection channels;
- determining the quality of the identified candidate site locations, wherein the quality of a candidate site location is determined based on relative intensities of fluorescent emission signals detected at the identified candidate site location by the different detection channels;
- and
- generating a template of nucleic acid site locations from the candidate site locations based on the quality of the identified candidate site locations, wherein identified candidate site locations that do not meet a threshold level of quality are excluded from the template.
A system for analysis of image data generated during nucleic acid sequencing, comprising a processer configured to execute instructions for performing a method comprising:
- obtaining fluorescent emission signals from nucleic acid sites on a flow cell, wherein a nucleic acid site comprises multiple copies of a nucleic acid, and wherein the fluorescent emission signals are indicative of nucleotides present at the nucleic acid site;
- identifying candidate site locations on the flow cell from the fluorescent emission signals, the fluorescent emission signals detected by different detection channels;
- determining the quality of the identified candidate site locations, wherein the quality of a candidate site location is determined based on relative intensities of fluorescent emission signals detected at the identified candidate site location by the different detection channels;
- and
- generating a template of nucleic acid site locations from the candidate site locations based on the quality of the identified candidate site locations, wherein identified candidate site locations that do not meet a threshold level of quality are excluded from the template.
A computer readable medium comprising instructions that when executed by a processor perform a method comprising:
- obtaining fluorescent emission signals from nucleic acid sites on a flow cell, wherein a nucleic acid site comprises multiple copies of a nucleic acid, and wherein the fluorescent emission signals are indicative of nucleotides present at the nucleic acid site;
- identifying candidate site locations on the flow cell from the fluorescent emission signals, the fluorescent emission signals detected by different detection channels;
- determining the quality of the identified candidate site locations, wherein the quality of a candidate site location is determined based on relative intensities of fluorescent emission signals detected at the identified candidate site location by the different detection channels;
- and
- generating a template of nucleic acid site locations from the candidate site locations based on the quality of the identified candidate site locations, wherein identified candidate site locations that do not meet a threshold level of quality are excluded from the template.
