Anti-inflammatory and analgesic compounds and use thereof

A compound has the structure shown in formula (I) or formula (II), or isomer and pharmaceutically acceptable salt thereof,

• wherein,
• R1, R2, R3, or R4 are independently hydrogen, deuterium, halogen, cyano, C1-3 alkoxy, hydroxyl, aldehyde group, carboxyl, C2-6 ester, amide, substituted amide, C1-3 alkyl, substituted C1-3 alkoxy, or substituted C1-3 alkyl, respectively, the said substituent is one or more selected from the group consisting of deuterium, halogen, hydroxyl, carboxyl, C1-3 alkyl or cyano;
• alternatively, two adjacent groups in R1, R2, R3 and R4 are joined together to form —O—R17—O— group, wherein R17 is C1-6 alkyl;
• R5, R6, R7 or R8 are independently hydrogen, deuterium, halogen, hydroxyl, cyano, carboxyl, C2-6 ester group or substituted or non-substituted groups of amino, amide, C1-3 alkoxy or C1-3 alkyl, the said substituent is one or more selected from the group consisting of deuterium, halogen, cyano, C1-3 alkyl or C1-3 haloalkyl;
• R9 is hydrogen, C1-4 alkyl, or C1-4 substituted alkyl, the said substituent is one or more selected from the group consisting of deuterium, hydroxyl, C1-2 alkoxy, or cyano;
• X is —C(═O)—, —S(═O)2—, —S(═O)—, —C(═NH)— or substituted —C(═NH)—, the substituent is selected from CN and SO2NH2;
• R10 is C1-4 alkyl, substituted C1-4 alkyl, C3-6 cycloalkyl, substituted C3-6 cycloalkyl, amino, C1-4 alkylamino, substituted C1-4 alkylamino, C3-6 cycloalkylamino, C1-4 alkoxy, substituted C1-4 alkoxy, or vinyl;
• or R9 is attached to R10 so that R9—N—X—R10 forms a ring group together;
• the said substituent is selected from one or more of deuterium, halogen and cyano;
• R11 is hydrogen, deuterium, halogen, cyano, C1-2 alkyl, C1-2 alkoxy, carboxyl, or C1-3 alkyl substituted amide;
• R12 is a hydrogen, deuterium, halogen, hydroxyl, cyano, carboxyl, or substituted or non-substituted group of the following:
• C1-4 alkyl, C3-6 cycloalkyl, C1-4 alkoxy, morpholinyl, aminocarbonyl, C1-4 alkylamino carbonyl, or C1-4 alkoxy carbonyl;
• the substituent is selected from one or more of deuterium, halogen or cyano;
• and
• (a) when R12 is hydrogen, R13 is cyano, aldehyde group, carboxyl, borono, substituted C1-4 alkoxy, or substituted or non-substituted group of C1-4 alkoxy carbonyl, amino carbonyl, phenyl amino carbonyl, pyridinyl amino carbonyl, C1-4 alkylamino carbonyl, C1-4 alkoxy carbonyl, C1-4 alkylamino carbonyl amino, C2-6 heterocycloalkyl carbonyl, C3-6 heterocycloalkyl ketone group, C1-2 alkyl carbonyl, sulfonamido, sulfonyloxy, amino sulfonyl, amino sulfonyloxy, C1-4 alkyl sulfonamido, C1-4 alkyl amino sulfonyl, C1-4 alkyl amino sulfonamido, C1-4 alkyl sulfonyloxy, C1-4 alkyl amino sulfonyloxy, C1-4 alkyl amino carbonyloxy, tetrazolyl, triazolyl, imidazolyl, morpholinyl, C1-4 alkyl thio, C3-6 cycloalkoxy or C3-6 heterocycloalkyloxy, the substituent is selected from one or more of deuterium, halogen, cyano, hydroxyl, phenyl, pyridyl, C1-3 alkyl, C1-3 deuteroalkyl or C1-3 haloalkyl;
• (b) when R12 is not hydrogen, R13 is cyano, aldehyde group, carboxyl, borono, or substituted or non-substituted group of C1-4 alkoxy carbonyl, amino carbonyl, phenyl amino carbonyl, pyridinyl amino carbonyl, C1-4 alkylamino carbonyl, C1-4 alkoxy carbonyl amino, C1-4 alkylamino carbonyl amino, C2-6 heterocycloalkyl carbonyl, C3-6 heterocycloalkyl ketone group, C1-4 alkyl carbonyl, sulfonamido, sulfonyloxy, sulfamoyl, sulfamoyloxy, C1-4 alkyl sulfonamido, C1-4 alkyl amino sulfonyl, C1-4 alkyl amino sulfonamido, C1-4 alkyl sulfonyloxy, C1-4 alkyl amino sulfonyloxy, C1-4 alkyl amino carbonyloxy, heterocyclic group, C1-4 alkyl amino, C3-6 cycloalkyl amino, C3-6 heterocycloalkyloxy, C3-6 heterocycloalkyl amino, C1-4 alkyl carbonyl amino, C1-4 alkoxy, C1-4 alkylthio, C3-6 cycloalkoxy, C3-6 heterocyclo alkyl or C1-4 alkyl, the substituent is selected from one or more of deuterium, halogen, cyano, hydroxyl, phenyl, pyridyl, C1-3 alkyl, C1-3 deuteroalkyl and C1-3 haloalkyl;
• R14 is a cyano, carboxyl, or substituted or non-substituted group of C1-4 alkoxy carbonyl, amino carbonyl, C1-4 alkyl amino carbonyl, C1-4 alkoxy carbonyl amino, C1-4 alkyl amino carbonyl amino, C2-6 heterocycloalkyl carbonyl, sulfamoyl, C1-4 alkylaminosulfonyl, heterocyclic, C3-6 heterocycloalkyl, C3-6 heterocycloalkyloxy, C1-4 alkoxy, C3-6 cycloalkoxy, C3-6 heterocycloalkoxy or C1-4 alkyl, the substituent is selected from one or more of deuterium, halogen, cyano, hydroxyl, C1-3 alkyl and C1-3 haloalkyl;
• R15 is hydrogen, deuterium, halogen, cyano, C1-2 alkyl, C1-2 alkoxy, carboxyl, or C1-3 alkyl substituted amide;
• provided that the following conditions are excluded:
• (i) In formula (I), when R1-R3 is methoxy, R4-R9 is hydrogen, R10 is methyl, R11-R12 is hydrogen, and R15 is hydrogen, R13 is selected from the following groups:
• C1-4 alkoxy, C1-4 alkylthio, deutero C1-4 alkylthio, C1-4-alkoxy carbonyl amino, butyl carbonyl, or morpholinyl;
• (ii) In formula (I), R1-R3 is methoxy, R4-R9 is hydrogen, R10 is deuteromethyl, R11 is hydrogen, R12 is deuterium, R13 is methoxy, and R15 is deuterium;
• (iii) In formula (I), R1-R3 is methoxy, R4-R9 is hydrogen, R10 is methyl, R11 is hydrogen, R12 is bromine, R13 is methoxy, and R15 is bromine.
