Abstract
The present invention relates to a microfluidic assay system and associated reading device, as well as the individual components themselves. The present invention also relates to methods of conducting assays, using a disposable system and associated reading device, as well as kits for conducting assays.

-65. (canceled)
A self-contained dry, planar microfluidic cartridge and an associated reader device, comprising:
- a sample input port for receiving a liquid sample to be assayed, the sample input port connected to a first microfluidic channel at a first end of said microfluidic channel;
- a second microfluidic channel fluidically connected to the first microfluidic channel at a first junction disposed at a second end of the first microfluidic channel;
- a plurality of assay channels each connected at a respective assay channel junction to the second microfluidic channel wherein at least one assay channel junction is disposed along the second microfluidic channel on a first side of the first junction and at least one assay channel junction is disposed along the second microfluidic channel on an opposite side of the first junction, each/said assay channel(s) comprises one or more reagents deposited therein for use in conducting an assay and a detection zone for use in detecting any analyte which may be present in a sample or analyte reaction product;
- and each/said assay channel(s) is in fluid communication with a compressible, gas-filled chamber downstream from each/said detection zone, wherein each/said gas-filled chamber lacks any valve or port that communicates with the outside of the strip, and wherein the self-contained dry planar microfluidic cartridge does not contain liquid prior to the application of a sample and comprises three resilient planar layers, which are sandwiched together to define each/said microfluidic channel(s) and gas-filled chambers and wherein said gas-filled chamber(s) are configured to be compressed, thereby causing the gas-filled chamber to be deformed and gas to be expelled from the gas-filled chamber(s) and upon decompressing the gas-filled chamber(s), the gas-filled chamber(s) are configured to return to its non-deformed state, and whereby gas is configured to be drawn back into the gas-filled chamber(s), wherein said compression and decompression in turn is configured to cause reciprocal movement of the liquid sample within said/each microfluidic channel.
1 more element
A method of conducting an assay on a liquid sample, the method comprising:
- a) compressing a/said gas filled chamber(s) of the self-contained dry planar microfluidic cartridge of claim 1, so as to expel gas from said/each gas filled chamber(s);
- b) allowing a liquid sample to be drawn by capillary action along the first microfluidic channel, into and along the first and opposite sides of the second microfluidic channel, and into and only partially along each said/each analysis channel(s);
- c) partially decompressing said/each gas filled chamber(s) thereby causing the gas to be drawn back into said/each gas-filled chamber(s) thereby causing the liquid sample disposed within each said/each assay channel(s) to be drawn further therealong;
- d) allowing one or more of the reagent(s) to react with any analyte present in the liquid sample;
- and
- e) detecting, within each said/each assay channels, analyte present therein.
