Abstract
This invention relates to compositions and methods for modifying root architecture in a plant through modification of endogenous DEEPER ROOTING 1 (DRO1) nucleic acids. The invention further relates to plants produced using the methods and compositions of the invention.
Claims28
A corn plant or part thereof comprising at least one mutation in an endogenous DEEPER ROOTING 1 (DRO1) gene encoding a DRO1 polypeptide, wherein the at least one mutation results in a C-terminal mutation in the DRO1 polypeptide, wherein the endogenous DRO1 gene:
- (a) comprises a nucleotide sequence having at least 80% sequence identity to any one of SEQ ID NOs:
- 69, 70, 72, 73, 75, or 76;
- (b) comprises a region having at least 80% sequence identity to any one of the nucleotide sequences of SEQ ID NOs:
- 78-81, 82-94, or 95-107;
- (c) encodes a polypeptide sequence having at least 80% identity to any one of the amino acid sequences of SEQ ID NOs:
- 71, 74 or 77 and/or
- (d) encodes a region having at least 80% identity to the amino acid sequence of SEQ ID NO:108 or SEQ ID NO:109.

A method for producing a corn plant or part thereof comprising at least one cell having a mutation in an endogenous DEEPER ROOTING 1 (DRO1) gene, the method comprising contacting a target site within the endogenous DRO1 gene in the corn plant or part thereof with a nuclease comprising a cleavage domain and a nucleic acid binding domain, wherein the nucleic acid binding domain of the nuclease binds to a target site within the endogenous DRO1 gene, wherein the endogenous DRO1 gene:
- (i) comprises a nucleotide sequence having at least 80% sequence identity to any one of SEQ ID NOs:
- 69, 70, 72, 73, 75, or 76;
- (ii) comprises a region having at least 80% sequence identity to any one of the nucleotide sequences of SEQ ID NOs:
- 78-81, 82-94, or 95-107;
- (iii) encodes a polypeptide having at least 80% identity to any one of SEQ ID NOs:
- 71, 74 or 77;
- and/or
- (iv) encodes a region having at least 80% identity to the amino acid sequence of SEQ ID NO:
- 108 or SEQ ID NO:109, thereby producing a corn plant or part thereof comprising at least one cell having a mutation in the endogenous DRO1 gene.

A guide nucleic acid that binds to a target site within an endogenous gene encoding DEEPER ROOTING 1 (DRO1), the endogenous gene:
- (a) comprises a nucleotide sequence having at least 80% sequence identity to any one of SEQ ID NOs:
- 69, 70, 72, 73, 75, or 76;
- (b) comprises a region having at least 80% sequence identity to any one of the nucleotide sequences of SEQ ID NOs:
- 78-81, 82-94, or 95-107;
- (c) encodes a polypeptide having at least 80% identity to any one of SEQ ID NOs:
- 71, 74 or 77;
- and/or
- (d) encodes a region having at least 80% identity to the amino acid sequence of SEQ ID NO:
- 108 or SEQ ID NO:109.
