Abstract
Provided herein are systems for increasing AAV particle production. These systems comprise producer cell lines adapted for the production of AAV particles, as well as methods of producing AAV particles using said producer cell lines. Also provided are AAV particles produced by said production systems, producer cell lines and methods.

A genetically engineered producer cell line in which the expression of at least one of TMED10, MON2, TMED2, HS2ST1, C3orf58, SPPL3, SURF4, LSM5, ARF1, and PI4 KB is reduced as compared to a control cell line, and/or in which the expression of at least one of B4GALT7, B3GAT3, OAF, EXT2, COMMD3, SLC35D1, B3GALT6, SDC1, NDST1, RAC1, CSK, GLCE, PDCL, FAM20B, TM4SF5, DGAT2, POMT1, YY1, and DPF2 is increased as compared to a control cell line.
A method for producing adeno-associated viral (AAV) particles comprising culturing a producer cell line in which the expression of at least one of TMED10, MON2, TMED2, HS2ST1, C3orf58, SPPL3, SURF4, LSM5, ARF1, and PI4 KB is reduced as compared to a control cell line, and/or in which the expression of at least one of B4GALT7, B3GAT3, OAF, EXT2, COMMD3, SLC35D1, B3GALT6, SDC1, NDST1, RAC1, CSK, GLCE, PDCL, FAM20B, TM4SF5, DGAT2, POMT1, YY1, and DPF2 is increased as compared to a control cell line, under condition allowing production and/or secretion of the AAV particles.
