Abstract
This invention relates to a device and method of using the device for purification that separates material of interest from contaminating materials using non-porous magnetic particles and single-use or disposable materials that come in contact with the material of interest. The process encompasses multiple cycles in a single batch to reduce the cost of magnetic particles. This method can be executed in a fully automated manner by a controller that manages different inputs and outputs of system hardware.
Claims19
A method of separating a material of interest from a starting material, the method comprising:
- (a) mixing the starting material with a plurality of non-porous magnetic particles to form a mixture, each of the non-porous magnetic particles having an outer surface with an affinity for the material of interest such that the material of interest binds to the non-porous magnetic particles;
- (b) subjecting the mixture to a magnetic field to capture the non-porous magnetic particles having the material of interest bound thereto;
- (c) washing the mixture with an equilibration buffer in the presence of the magnetic field to remove any starting material not bound to the non-porous magnetic particles;
- and
- (d) mixing the non-porous magnetic particles with an elution buffer in the absence of the magnetic field to elute the material of interest from the non-porous magnetic particles.
9 The method according to claim 1, wherein the plurality of non-porous magnetic particles is regenerated by exposing the plurality of non-porous magnetic particles to a regeneration buffer in the absence of the magnetic field followed by exposure to the regeneration buffer in the presence of magnetic field to remove any non-specific impurities bound to each of the non-porous magnetic particles.
A method of separating a material of interest from a starting material, the method comprising:
- (a) mixing the starting material with a plurality of non-porous magnetic particles to form a mixture, each of the non-porous magnetic particles having an outer surface with an affinity for the material of interest such that the material of interest binds to the non-porous magnetic particles;
- (b) subjecting the mixture to a magnetic field to capture the non-porous magnetic particles having the material of interest bound thereto;
- (c) washing the captured non-porous magnetic particles with an equilibration buffer in the presence of the magnetic field to remove any remaining starting material not bound to the non-porous magnetic particles;
- (d) mixing the non-porous magnetic particles with an elution buffer in the absence of the magnetic field to elute the material of interest from the non-porous magnetic particles;
- (e) recirculating the equilibration buffer in the absence of magnetic field, and then in the presence of the magnetic field to further wash the non-porous magnetic particles;
- and
- (f) repeating steps (c) and (d), thereby improving separation of the material of interest from the starting material.
A method of separating a material of interest from a starting material, the method comprising:
- (a) providing a plurality of non-porous magnetic particles into a separation chamber, each of the non-porous magnetic particles having a cubic or tetrahedron shape and an outer surface with an affinity for the material of interest;
- (b) washing the plurality of non-porous magnetic particles with an equilibration buffer in the presence of a magnetic field;
- (c) mixing the starting material with the plurality of non-porous magnetic particles to form a mixture in the separation chamber in which the material of interest binds to the non-porous magnetic particles;
- (d) subjecting the mixture to a magnetic field to capture the non-porous magnetic particles having the material of interest bound thereto;
- (e) introducing a regeneration buffer to the mixture in the absence of the magnetic field followed by exposure to the regeneration buffer in the presence of the magnetic field to remove any unwanted impurities bound to each of the non-porous magnetic particles;
- (f) washing the plurality of non-porous magnetic particles with an equilibration buffer in the presence of the magnetic field;
- and
- (g) repeating steps (e) and (f) one or more times, thereby improving separation of the material of interest from the starting material.
