Abstract
There is provided amino acid cleaving reagents with improved cleavage activity, allowing for more structural information to be obtained from polypeptides in sequencing reactions.

A composition comprising:
- a first cleaving reagent comprising a first aminopeptidase from Pyrococcus horikoshii and a first tag sequence;
- and
- a second cleaving reagent comprising a second aminopeptidase from Pyrococcus horikoshii,
- wherein the first and second aminopeptidases have different amino acid sequences.

- 1;
- and
- a third cleaving reagent comprising a third aminopeptidase having an amino acid sequence that is at least 80% identical to SEQ ID NO:
- 5 or 7.
