Abstract
Functionalized aryldiazonium salts and films formed by electrografting of functionalized aryldiazonium salts are provided. Methods for purifying functionalized aryldiazonium salts and for coating solid support systems with functionalized aryldiazonium salts are also provided. These coated solid support systems can be used, for example, in methods of oligonucleotide synthesis.
Claims64
A purified aryldiazonium tetrafluoroborate salt.
A method for preparing a purified aryldiazonium tetrafluoroborate salt comprising:
- a) dissolving a starting aniline in ethanol and adding boron trifluoride-THF complex;
- b) cooling the resulting solution and adding t-butylnitrite;
- c) warming the resulting solution to room temperature;
- and
- d) collecting the precipitated salts directly or as oils.
A solid support system for synthesis of oligonucleotides, wherein the support is an electrode coated with a thin film comprising functional groups, wherein the electrode is a platinum electrode, a titanium electrode, or a titanium nitride electrode.
A solid support system for synthesis of oligonucleotides, wherein the support is an electrode coated with a thin film comprising polybenzyl alcohol.
A method of coating a solid support system for synthesis of oligonucleotides with a thin film comprising a functional group, the method comprising treating the solid support with a solution comprising an aryldiazonium salt and optionally an electrolyte and optionally energizing the solid support, thereby producing a coated solid support.
A method of preparing an electrode surface for oligonucleotide synthesis, comprising
- (a) cleaning the electrode surface,
- (b) introducing a diazonium salt to the cleaned electrode surface,
- (c) electrografting the diazonium salt so as to form a multi-layer film of polyalcohol onto the cleaned electrode surface,
- wherein the multi-layer film comprises a loose outer layer of polyalcohol and an inner stable layer of polyalcohol,
- (d) stripping the loose outer layer of polyalcohol, and
- (e) performing the oligonucleotide synthesis on the inner stable layer polyalcohol.
