Abstract
Compositions for producing glycoengineered proteins, e.g. antibodies, include host cells which lack the ability to produce enzymes that modulate sialic acid metabolic flux.
Claims24
A method of producing modified host cells for producing engineered proteins or peptides comprising:
- transfecting host cells with an expression vector encoding a protein or peptide comprising one or more genetic mutations, wherein the one or more genetic mutations encode for one or more N-glycans;
- thereby, producing a modified protein peptide.
16-27. (canceled)
A modified host cell comprising one or more nucleic acid sequences encoding one or more enzymes that modulate N-glycan branching, sialylation or combinations thereof.
29-36. (canceled)
An engineered protein comprising one or more modified amino acid sequences, wherein the modified amino acid sequences comprise an N-glycan consensus sequence.
38-47. (canceled)
A modified immunoglobulin comprising one or more N-glycan groups.
51-54. (canceled)
