Abstract
Provided are new bifunctional chelators for use in the manufacture of radiolabeled targeting agents for therapeutic or diagnostic use. Also provided are conjugates of the new bifunctional chelators with various cancer antigen targeting agents.

A bifunctional chelator compound comprising a formula
- wherein R is a reactive group;
- wherein L1 comprises a formula
- wherein:
- R2 is —C(S)—, —NHC(S)—, —CH2CH2C(O)—, and C(O)CH2CH2C(O)—;
- R3 is CH2, CH2NH, or (CH2)n1, where n1 is an integer from 1 to 5;
- R4 is a cyclic alkane having 5 to 8 carbons or linear alkane having 1 to 6 carbons;
- and
- R5 is an amino acid or amino acid derivative;
- wherein n is an integer selected from the group consisting of 1, 2, and 3;
- and
- wherein each of n2, n3, n4, n5, no, n7, and ng is independently 0 or 1;
- and
- wherein M is a chelator moiety.
