Abstract
Aspects of the present disclosure relate to methods and compositions for genetic modification of immune cells to generate immune cells that do not secrete TGF-β1. Also disclosed are methods for use of such genetically modified immune cells, including immunotherapeutic methods for the treatment of cancer. Further aspects of the disclosure are directed to human CD8+ cells comprising a genetic modification that prevents the cells from secreting TGF-β1.
Claims87
A method for treating a subject having cancer, the method comprising administering to the subject a human CD8+ T cell that does not secrete TGF-β1.
A method for genetically modifying a human CD8+ T cell, the method comprising introducing into the cell a Cas nuclease and a guide RNA targeted to TGFB1.
A method for treating a subject having cancer, the method comprising:
- (a) modifying immune cells from the subject to generate genetically modified immune cells that do not secrete TGF-β1;
- and
- (b) administering the genetically modified immune cells to the subject.
A human immune cell comprising a genetic modification that prevents the cell from secreting TGF-β1.
