Abstract
Provided herein are methods and compositions for the treatment of cancer by modulating the microbiome to enhance the efficacy of immune checkpoint blockade. The microbiome may be modulated by the administration of butyrate and/or butyrate-producing bacteria. Also provided herein are methods of determining a response to an immune checkpoint inhibitor by identifying if a subject has a favorable microbial profile.

A method of reducing or delaying the growth of a tumor in a human subject in need thereof, comprising administering to the subject's gastrointestinal tract a composition comprising bacteria belonging to the family Ruminococcaceae in an amount effective to increase relative abundance of bacteria belonging to the family Ruminococcaceae in the subject's gut.
A method of treating a cancer in a human subject in need thereof, comprising
- a) administering to the subject's gastrointestinal tract a composition comprising bacteria belonging to the family Ruminococcaceae in an amount effective to increase relative abundance of bacteria belonging to the family Ruminococcaceae in the subject's gut, and
- b) administering to the human subject an effective amount of a human programmed cell death 1 (PD-1) axis binding antagonist.
A method of treating metastatic melanoma in a human subject in need thereof, comprising
- a) orally administering to the subject a composition comprising bacteria belonging to the family Ruminococcaceae in an amount effective to increase relative abundance of bacteria belonging to the family Ruminococcaceae in the subject's gut, and
- b) intravenously administering to the subject an effective amount of pembrolizumab.
