Abstract
A method of analyzing a molecule in a nanopore is disclosed. A voltage is applied across a nanopore that is inserted in a membrane by coupling the nanopore to a voltage source. The nanopore is decoupled from the voltage source. After the decoupling, a rate of decay of the voltage across the nanopore is determined. A molecule in the nanopore is distinguished from other possible molecules based on the determined rate of decay of the voltage across the nanopore.
Claims14
A system for analyzing a molecule in a nanopore, the system comprising:
- a well having an opening configured to support a membrane having a nanopore disposed therein;
- a working electrode disposed within the well;
- a counter electrode located outside the well such that the membrane, when formed over the opening of the well, is located between the working electrode and the counter electrode;
- a voltage source in electrical communication with the working electrode, wherein the working electrode in connection with the voltage source and counter electrode are configured to apply a voltage between the working electrode and the counter electrode;
- a measuring circuit configured to measure a change in a voltage across the nanopore;
- and
- a processor configured to distinguish a molecule in the nanopore from other possible molecules based at least in part on the measured change in the voltage across the nanopore.
