Abstract
The present disclosure provides methods and compositions for sequencing nucleic acid molecules and identifying individual sample nucleic acid molecules using Molecular Index Tags (MITs). Furthermore, reaction mixtures, kits, and adapter libraries are provided.
Claims18
A composition comprising:
- at least 1,000,000 tagged sample nucleic acid molecules, wherein each of the tagged sample nucleic acid molecules comprises (a) a sample nucleic acid segment between 10 and 1,000 nucleotides in length linked to (b) at least one heterologous adaptor segment comprising a Molecular Index Tag (MIT) between 4 and 8 nucleotides in length;
- wherein the number of different MITs in the composition is between 10 and 3,000;
- wherein a ratio of the number of tagged sample nucleic acid molecules in the composition to the number of different MITs is at least 1,000:1;
- wherein the sequences of different MITs differ from each other by at least 2 nucleotides;
- and wherein the composition comprises at least two copies of each MIT.
