Abstract
This invention relates to compounds, pharmaceutical compositions comprising them, and methods of using the compounds and compositions for treating diseases related to translesion synthesis (TLS) pathway. More particularly, this disclosure relates to small molecule inhibitors of TLS, methods of inhibiting TLS pathway with these compounds, and methods of treating diseases related to the TLS pathway.
Claims29
A method of treating cancer, comprising administering to a subject in need thereof (i) one or more secondary therapeutic agents and (ii) one or more compounds of formula (I),
- or a pharmaceutically acceptable salt thereof, wherein
- X and Y are independently selected from NR, O, or S,
- where R is hydrogen or C1-C4 alkyl;
- R1, R2, R3, and R4 are independently selected from the group consisting of hydrogen, halogen, —NO2, —CN, C1-C6 alkyl, C1-C6 haloalkyl, —NH2, —NH(C1-C6 alkyl), —N(C1-C6 alkyl)2, —OH, C1-C6 alkoxy, C1-C6 haloalkoxy, —SH, hydroxy(C1-C6 alkyl), alkoxy(C1-C6 alkyl), amino(C1-C6 alkyl), —CONH2, —CONH(C1-C6 alkyl), —CON(C1-C6 alkyl)2, —CO2H, —CO2(C1-C6 alkyl), —CHO, —CO(C1-C6 alkyl), and —S(O)0-2(C1-C6 alkyl);
- R5, R6, and R7 are independently selected from the group consisting of hydrogen, halogen, —NO2, —CN, C1-C6 alkyl, C1-C6 haloalkyl, —NH2, —NH(C1-C6 alkyl), —N(C1-C6 alkyl)2, —OH, C1-C6 alkoxy, C1-C6 haloalkoxy, —SH, —CONH2, —CONH(C1-C6 alkyl), —CON(C1-C6 alkyl)2, —CO2H, —CO2(C1-C6 alkyl), —CHO, —CO(C1-C6 alkyl), and —S(O)0-2(C1-C6 alkyl),
- or R5 and R6, together with the atoms to which they are attached, form a 5 or 6 membered aryl, heteroaryl, or heterocyclyl;
- R8 is C1-C8 alkyl optionally substituted with one or more R10, C2-C8 alkenyl optionally substituted with one or more R10, or C2-C8 alkynyl optionally substituted with one or more R10,
- wherein each R10 is independently selected from the group consisting of halogen, —NO2, —CN, —NH2, —NH(C1-C6 alkyl), —N(C1-C6 alkyl)2, —OH, C1-C6 alkoxy, C1-C6 haloalkoxy, —CONH2, —CONH(C1-C6 alkyl), —CON(C1-C6 alkyl)2, —CO2H, —CO2(C1-C6 alkyl), —CHO, —CO(C1-C6 alkyl), and —S(O)0-2(C1-C6 alkyl), or two R10 groups when attached to the same carbon atom form ═O, ═NR, or ═N—OH;
- and
- R9 is hydrogen or C1-C4 alkyl.
