Abstract
Embodiments of the present disclosure include methods and compositions for functionalizing molecules, such as oligonucleotides, with functional groups, including polyhistidine tags useful in affinity methods. Some embodiments include methods for modifying and purifying complex mixtures of molecules by exchange of functional tags.
Claims19
A method of making functionalized-oligonucleotides, comprising:
- providing at least two sets of oligonucleotides, a first set of oligonucleotides comprising a 5′-phosphate and a 3′-terminal block, and a second set of oligonucleotides comprising a 5′-functional group, wherein the 5′-functional group is an aldehyde group or comprises a biotin moiety;
- ligating an oligonucleotide from the first set of oligonucleotides with an oligonucleotide from the second set of oligonucleotides to form functionalized-oligonucleotides.
