Abstract
The present application provides single-domain antibodies targeting CD33 and constructs thereof, including chimeric receptors, immune effector cell engagers and immunoconjugates. Further provided are engineered immune cells (such as T cells) comprising an anti-CD33 chimeric receptor and optionally a second chimeric receptor targeting a second antigen or epitope. Pharmaceutical compositions, kits and methods of treating cancer are also provided.

An anti-CD33 construct comprising a single domain antibody (“sdAb”) moiety that specifically binds to CD33, wherein the sdAb moiety comprises a CDR1 comprising the amino acid sequence of any one of SEQ ID NOs:
- 4, 11, 18, 25, 32, 39, 46, 53, 60, 67, 74, 81, 88, 95, and 102, or a variant thereof comprising up to about 3 amino acid substitutions in the CDR1;
- a CDR2 comprising the amino acid sequence of any one of SEQ ID NOs:
- 6, 13, 20, 27, 34, 41, 48, 55, 62, 69, 76, 83, 90, 97, and 104, or a variant thereof comprising up to about 3 amino acid substitutions in the CDR2;
- and a CDR3 comprising the amino acid sequence of any one of SEQ ID NOs:
- 8, 15, 22, 29, 36, 43, 50, 57, 64, 71, 78, 85, 92, 99, and 106, or a variant thereof comprising up to about 3 amino acid substitutions in the CDR3.
