Abstract
Provided are compounds of Formula (I): and pharmaceutically acceptable salts and compostions thereof, which are useful for treating a variety of conditions associated with methyl modifying enzymes.
Claims28
A compound having the Formula I:
- or a pharmaceutically acceptable salt thereof, wherein
- each X is independently CH or N;
- R1 is (C1-C4)alkyl, —(C3-C5)cycloalkyl, or (C1-C4)haloalkyl;
- R2 is hydrogen, halo, (C1-C4)alkyl, halo(C1-C4)alkyl, (C1-C4alkoxy, halo(C1-C4alkoxy, or —CN;
- R3 is (C1-C4)alkyl or halo(C1-C4)alkyl;
- R4 is (C1-C4)alkyl, halo(C1-C4)alkyl, halo(C1-C4)alkoxy, —(C1-C4)alkylOH, —(C1-C4)alkylO(C1-C4)alkyl, phenyl, 5-6 membered heteroaryl, monocyclic (C3-C7)cycloalkyl, bridged bicyclic (C5-C8)cycloalkyl, or 4-7 membered monocyclic heterocyclyl, wherein said phenyl, heteroaryl, monocyclic cycloalkyl, bridged bicyclic cycloalkyl, and heterocyclyl are each optionally substituted with 1 to 3 groups selected from R5;
- R5 is selected from (C1-C6)alkyl, (C3-C6)cycloalkyl, halo(C3-C6)cycloalkyl, —(C1-C4)alkylOH, halo(C1-C4)alkyl, (C1-C4)alkoxy, halo(C1-C4)alkoxy, halo, nitrile, amide, urea, carbamate, sulfonamide, hydroxyl, oxo, OR6, —OC(O)(C1-C4)alkyl, —C(O)(C1-C4)alkyl, —C(O)(C1-C4)alkylOH, —C(O)(C1-C4)alkylNH(C1-C4)alkyl, —C(O)(C1-C4)alkylN((C1-C4)alkyl)2, —C(O)(C1-C4)alkylNH2, —SO2(C1-C4)alkyl, (C1-C4)alkyl-O—(C1-C4)alkyl, substituted phenyl, 5-6 membered heteroaryl, 4-7 membered heterocyclyl, and NRARB, wherein said cycloalkyl for R5 is optionally substituted with —O(C1-C4)alkyl;
- RA is hydrogen, (C1-C4)alkyl, halo(C1-C4)alkyl, (C3-C6)cycloalkyl, 4-7 membered heterocyclyl, or (C1-C4)alkyl-O—(C1-C4)alkyl;
- RB is hydrogen, (C1-C4)alkyl, —(C3-C5)cycloalkyl, or 4-7 membered heterocyclyl, wherein said (C1-C4)alkyl for RB is optionally substituted with 1 to 3 groups selected from halo, phenyl, (C1-C4)alkyl, (C1-C4)alkoxy, and OH and wherein said heterocyclyl, —(C3-C5)cycloalkyl for RB is optionally substituted with 1 to 3 groups selected from (C1-C4)alkyl, halo(C1-C4)alkyl, hydroxy(C1-C4)alkyl, —O(C1-C4)alkyl, and —C(═O)O(C1-C4)alkyl;
- or
- RA and RB taken together with the nitrogen atom to which they are attached form a 3-7 membered heterocyclyl optionally substituted with 1 to 3 groups selected from halo, (C1-C4)alkyl, halo(C1-C4)alkyl, (C1-C4)alkoxy, oxo, halo(C1-C4)alkoxy, 5-6 membered heteroaryl, (C3-C6)cycloalkyl, —SO2(C1-C4)alkyl, NMe2, hydroxy, cyano, spirooxetanyl and (C1-C4)cycloalkoxy optionally substituted with (C1-C4)alkyl, halo(C1-C4)alkyl, or (C1-C4)alkoxy;
- and
